Evaluation of DNA Isolated from Stored Bone Marrow Slides




Abstract
Moleculalr analysis is the powerful tool for the diagnosis and research of various diseases. Because fresh or cryopreserved cell and tissue, which are the most suitaible samples for molecular analysis, may not always available, we investigate the
usefulness of DNA isolation from stored bone marrow slides as a source of cellular material. DNA amounts per slide varied from 3§¶ to 30§¶(mean 13§¶). High molecular weight(HMW) DNA was isolated from samples sored up to 2 years, and samples
stored
over
3 years showed some degradation of HMW DNA. Southern blot showed strong hybrdized bands in each sample stored up to 3 years, and PCR results showed excellent bands in all smaples for the 5 years. Stored bone marrow slides are very good source for
obtaining cellular material. Especially for the molecular analysis of rare disease and follow-up of patient.
